What about Witches or Warped Vines from The Warped Forest in the Nether? I'm thinking that Sculks are part of The Warped Forest since they have simular colors to them. Perhaps the witches took the Warped Viness and did some magic with them. This might explain the souls/soulsand that can also be found in the nether. SInce both Warped Viness and Soulsand are from the same biome, the witch might've used this combo with a certain potion to make the Warden. Going back on what you said on the Iron Golems, perhaps the witches used a spare Golem from the Illagers to test out this experiment. This might also explain why they lerk in the new caves that are filled with these new plants, since sculks just might be Warped Vines that are consumed by the souls that are from the soulsand. The warden looks like a Ravenger more than a Iron Golem however, and since Ravengers are slightly taller than Iron Golems (If they stood on their back legs only), the witches probably used one of them for that experiment. Ravengers are gray, but slightly green, combined with Warped Vines and the souls, you have a plant like Ravenger. Ravengers have those horns, so what if the Warped Vines changed its body and made it's horns sculks. These souls from the soulsand (and perhaps the burning rage of Soulfire) might explain why you can hear a heartbeat from the Warden, since the Ravenger would have a heart and the Iron Golem would not.
